Study on Maturation of Human Immature Oocyte in Vitro


Abstract
In mammals the preovulatory surge of gonadotropins initiates a series of events that lead to the maturation and subsequent ovulation of one or more oocytes. The occurrence of a complex sequence of maturational changes during the process of
follicle
growth confers on the oocytes the capacity to undergo normal fertilization and subsequent embryonic development.
To establish the possibility and extent on maturation of human immature oocyte in vitro without hormonal stimulation, and to confirm the influence of hyaluronidase on oocyte maturation with in vitro, the follow study was performed.
The study was designed as 3 groups; immature oocytes were treated by hylauronidase(Group I, n=122), not treated by hyaluronicase(Group II, n=92), and obtained from IVF program after controlled hypersimulation(Group III, n=44).
@ES The results were as below:
@EN 1) Extrusion rate of 1 st polar body without superovulation was 5.24¡¾4.3%.
2) After 12 hours of culture in vitro, the extrusion rate of 1st polar body and the rate of germinal vesical break down(GVBD) IN Group I, Group II, and Group III were(14.51¡¾4.40%, 15.58¡¾9.915, (17.25¡¾7.16%, 25.0¡¾8.66%) and (15.42¡¾3.72,
22.34¡¾9.84%) respectively.
3) After 24 hours of culture in vitro, the extrusion rate of 1st polar body and the rate of 1 st polarbody and the rate of GVBD in Group I, group II, and Group III were (32.36¡¾7.40%, 26.63¡¾8.93%), (35.11¡¾6.25%, 29.71¡¾7.10%) and (36.24¡¾7.4%,
31.23¡¾6.94%) respectively.
4) In Group I & II, the cleavage rate after 24 hours culture was 38.505 and after 48 hours culture, the cleavage rate was 57.47%.
5) In Group I & II, after 24 hours and 48 hours of culture, the cleavage rate was 28.505, 57.47% respectively. In Group III, after 24 hours and 48 hours of culture, cleavage rate was 22.72%, 47.72% respectively.
In conclusion, the human immature oocyte I vitro could be matured without the hormonal stimulation. Therefore, these immature oocytes from IVF program could be matured readily which should increased the oppotunity for securing a pregnancy
clinically.
